A Method for chromosome location of new Autosomal recessive mutations of Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) based on crosses with three strains bearing autosomal-Y translocations Neotropical Entomology

A method is proposed to locate any new autosomal and recessive mutation in the Mediterranean fruit fly, Ceratitis capitata (Wied.). A series of crosses searches for pseudo-linkage between sex and the mutation by employing three strains with Y-linked translocations, thereby indicating its chromosome location.

Cytogenetic and molecular analysis of the Pantaneiro cattle breed PAB

The objective of this work was to characterize Pantaneiro cattle genetically through its paternal ancestry by the morphology of the Y chromosome, whether submetacentric or acrocentric, as well as to identify the maternal ancestry through mitochondrial DNA. The karyotype and mitochondrial DNA of 12 bulls of Pantaneiro breed were analyzed. The Y chromosome was analyzed in lymphocyte metaphases and the mitochondrial DNA by diagnosing its haplotype (Bos taurus and Bos indicus). Among Pantaneiro animals analyzed three had a taurine (submetacentric) Y and nine had a zebuine (acrocentric) Y chromosome, suggesting breed contamination by Zebu cattle, once Pantaneiro is considered to be of European origin. ABSTRACT A total of 80 samples were collected to analyze the mitochondrial DNA (mtDNA) and Y chromosome genetic diversity of two main Portuguese Lidia bovine populations and clarify their genetic relationships with Spanish Lidia lineages. A 521-bp D-loop fragment was sequenced in 40 animals belonging to four herds (three Brava dos Açores and one Casta Portuguesa) and the same number of samples were collected with equal distribution from males in order to genotype six microsatellites, one single nucleotide polymorphism, and one indel. The mtDNA diversity recorded was similar to that observed in Lidia cattle. Prevalence of Y chromosome deletions in a Brazilian population of nonobstructive azoospermic and severely oligozoospermic men BJMBR

We determined the prevalence of Y chromosome deletions in a population of 60 Brazilian nonobstructive azoospermic and severely oligozoospermic men. PCR-based screening of microdeletions was performed on lymphocyte DNA for the presence of 14 sequence-tagged sites (STS) located in the azoospermic factor (AZF) on the Yq chromosome. All STS were amplified efficiently in samples from 12 fertile men tested, but failed to be amplified in samples from fertile women, indicating the specificity of PCR conditions for Yq screening. Reproductive deficiency in bulls from synthetic breeds according to the type of crossbreed and the morphology of the Y chromosome Genet. Mol. Biol.

We used 550 Braford and 214 Brangus-Ibagé bulls to study the association between Y chromosome morphology and the evaluation of breeding-soundness, karyotyping and breeding records were used to assess Y chromosome morphology. In general, no direct association was detected between the individual type of Y chromosome and reproductive fitness as previously estimated through breeding soundness examination. The type of breeding strategy used to form the synthetic breeds was also analyzed. The Braford breed is about 3/8 Zebu (Bos taurus indicus) and 5/8 Hereford (Bos taurus taurus) but when the breeding strategy used 1/4 Zebu animals to produce Braford bulls which were 3/8 Zebu a greater percentage of the bulls had to be culled due to reproductive problems. Y chromosomal STRs show sufficient variability among individduals in a population and a high degree of geographical differentiation, such that their polymorphic character makes them especially suited for population genetic studies. To investígate the polymorphism of a set of 17 Y-STR loci in northern China, we genotyped the 17 Y chromosomal STR loci in a population sample of 377 unrelated males from eight ethnic populations in northern China.
